چکیده مقاله:
p53 is a key tumor suppressor gene that is targeted for inactivation during human tumorigenesis. In this study, we produced and characterized polyclonal antihuman p53 antibody. The cDNA encoding the completehuman p53 protein was cloned into pGEX-4T-1 and expressed in Escherichia coli as a fusion protein with Schistosoma japonicum glutathione S-transferase (GST). The rabbits were immunized with the purifiedp53 recombinant protein. The obtained antisera were purified to increase the specificity of recognition. Thesensitivity and specificity of the produced antibody was analyzed by enzyme-linked immunosorbent, immunoblot, immunofluorescence and chromatin immunoprecipitation assays. Enzyme-linked immunosorbent assay showed that immunization with purified GST-p53 produced the high titer (1:10000) polyclonal antibodies with high specificity. Anti-p53 antibody allowed the sensitive detection of native p53 protein in immunoblotting, immunofluorescence and chromatin immunoprecipitation assays. Our results showed that anti-GST-p53 antibody provides a good means for studying the p53 expression pattern and its binding ability to other proteins in tumors.

Specific antibody immune response against the parasitic portion of a glutathione-S-transferase fusion protein.
The humoral immune response against an Entamoeba histolytica recombinant protein has been investigated. The 628 bp Bam HI-Eco RI DNA fragment (L1b) from the M11 cDNA clone, partially coding for a 220 kDa (L220) protein, was ligated in-frame into the pGEX-3X plasmid vector to produce the fusion protein GST-L1b. BALB/c mice were immunized with different doses of the GST-L1b fusion protein (10-500...

متن کاملMelittin-glutathione S-transferase fusion protein exhibits anti-inflammatory properties and minimal toxicity.
Although potent, proteins often require chemical modification for therapeutic use. Immunogenicity, difficult synthesis, and scale-up of these modifications are all engineering obstacles that stand in the way of expanding the use of these therapeutics. Melittin, a peptide derived from bee venom, has been shown to modulate inflammation. Although potentially therapeutic, the native peptide causes ...

Polyclonal antibodies against kappa light chain are used to diagnose diseases producing free light chain. The kappa and lambda light chains are products of immunoglobulin synthesis and released into the circulation in minor amounts such as serum, cerebrospinal fluid, urine and synovial fluid in normal condition. The purpose of this study was the production and purification of polyclonal immunog...
